    The role of ‘DELLA’ proteins in alternative splicing

    DELLA proteins, the master repressors of the gibberellin (GA) signaling pathway, are thought to regulate gene expression by mechanisms other than their interaction with bona fide transcription factors. In particular, current work in our lab shows that DELLA and their atypical interactors prefoldins are able to interact with and regulate a number of proteins that are part of the basal transcriptional machinery, regulating the RNA polymerase II activity. Among the novel interactors being studied are proteins involved in mRNA splicing. In this TFM, we have investigated the participation of DELLA and prefoldin in alternative splicing by analyzing protein-protein interaction between these proteins and Sm/LSm, which form heteroheptameric complexes and are an essential part of the spliceosome. We have also advanced in obtaining resources to test genetic interactions between della and sm/lsm mutants. In addition, some alternative splicing events affected by DELLA activity identified in a transcriptomic analysis by RNA-seq, have been validated by semiquantitative RT-PCR. Finally, it has been shown that GA signaling alters the Sm and LSm protein levels.En el laboratorio se está investigando los mecanismos moleculares por los que las proteínas DELLA regulan la expresión génica en plantas. El mecanismo aceptado en la actualidad indica que esta regulación ocurre a través de la interacción de las DELLA con numerosos factores de transcripción. No obstante, resultados recientes del laboratorio apuntan a mecanismos alternativos de regulación génica por parte de estas proteínas. Una de las claves fue la identificación de la interacción de las DELLA con el complejo prefoldina. Este complejo heterohexamérico (PFD1-6) está conservado de levaduras a humanos y se aisló originalmente gracias a su papel en el citoplasma, donde ejerce de co-chaperona ayudando en el plegamiento de tubulinas. En el laboratorio se ha demostrado que las DELLA promueven la acumulación en el núcleo del complejo y los últimos resultados apuntan a que puede tener un papel relevante en la regulación génica. En particular, hemos demostrado que tanto las DELLA como varias subunidades prefoldina interaccionan con varias proteínas Sm y LSM. Las proteínas Sm y LSM forman las 5 snRNP (del inglés, small nuclear ribonucleoprotein particles) del espliceosoma junto con UsnRNA (RNA pequeños nucleares ricos en uridina). Por un lado, las proteínas LSM forman dos tipos de heteroheptámeros (LSM1-7 y LSM2-8), siendo el primero citosólico y participando en el decaimiento de los mRNA mientras que el segundo se localiza en el núcleo estabilizando al snRNA U6. Por otro lado, las otras 4 snRNP, están formadas por un anillo de 7 proteínas Sm (B, D1, D2, D3, E, F y G) y 4 snRNA diferentes (U1, U2, U4 y U5). Además, se ha demostrado que la pérdida de función de PFD provoca alteración en los niveles de U6. Mediante un RNAseq se comprobó que los mutantes pfd4 mostraban fenómenos de procesamiento alternativo de mensajeros (splicing alternativo) diferentes al silvestre tras un tratamiento de frío. Por lo tanto, proponemos que las DELLA pueden tener un rol directo en el procesamiento alternativo. En el contexto de este TFM vamos a evaluar el efecto de DELLA y prefoldinas en el procesamiento alternativo de mensajeros mediante la comprobación de la interacción proteína-proteína entre DELLA y Sm/LSM, y PFD y Sm; tanto por técnicas de doble híbrido como Co-IP. De igual manera, se comprobarán las interacciones genéticas entre mutantes della y sm/lsm. También se confirmarán dianas de splicing alternativo, obtenidas mediante un RNAseq, con PCR cuantitativa o semi-cuantitativa. Finalmente se estudiará si las giberelinas alteran la localización y/o la configuración de los complejos Sm y/o LSM2-8 mediante análisis en el microscopio confocal y cromatografía de exclusión molecular, respectivamente. Los análisis se realizarán en varios fondos mutantes y líneas transgénicas que tienen niveles alterados de DELLA, y en una serie de mutantes pfd preparados en el laboratorio, de manera que podremos diseccionar genéticamente la contribución de las DELLA y PFD.Calleja Cabrera, J.     Video over DSL with LDGM Codes for Interactive Applications

    Digital Subscriber Line (DSL) network access is subject to error bursts, which, for interactive video, can introduce unacceptable latencies if video packets need to be re-sent. If the video packets are protected against errors with Forward Error Correction (FEC), calculation of the application-layer channel codes themselves may also introduce additional latency. This paper proposes Low-Density Generator Matrix (LDGM) codes rather than other popular codes because they are more suitable for interactive video streaming, not only for their computational simplicity but also for their licensing advantage. The paper demonstrates that a reduction of up to 4 dB in video distortion is achievable with LDGM Application Layer (AL) FEC. In addition, an extension to the LDGM scheme is demonstrated, which works by rearranging the columns of the parity check matrix so as to make it even more resilient to burst errors.     The purpose of any territorial strategic planning process, in its urban or environmental component, is to influence the future dynamics of the city and its area of influence by making coordinated and consensual decisions, that will be carried out by political, economic and social agents. Strategic planning is the key to achieving a 21st century city, according to the Challenges of the New Urban Agenda (III Habitat Conference of the United Nations, Quito, 2016). These challenges are to plan a city with this multiple approach: Compact, Inclusive, Connected and Resilient. In this sense, this study makes a methodological proposal for the development of a Strategic Plan as an instrument capable of merging and articulating the physical planning of the land (urban, urbanizable, rustic and protected), with the socio-economic strategies of the city, so that the development was matched to the requirements and needs of each moment, given the flexibility of this tool to design new scenarios, such as the selected case of Mérida, autonomous capital of Extremadura (Spain).info:eu-repo/semantics/publishedVersio

    Relación Entre La Gestión Del Cambio Y La Identificación Institucional De Los Servidores Públicos Del Hospital Belén De Trujillo, 2018

    La presente investigación se desarrolló con el objetivo de determinar la relación entre la gestión del cambio y la identificación institucional de los servidores públicos del Hospital Belén de Trujillo, 2018. Investigación de enfoque cuantitativo, de tipo no experimental, diseño correlacional, transversal, la población fueron los servidores públicos de la parte administrativa del Hospital Belén de Trujillo, 2018, la muestra fue universal por tener una población pequeña; el método utilizado fue el hipotético deductivo, la técnica fue la encuesta y para la recolección de datos se utilizó dos cuestionarios tipo escala Likert, los mismos que fueron validados con el coeficiente de V de Aiken, y la confiabilidad con el Alfa de Cronbach, para gestión del cambio a = 0. 979 (Excelente), y para identificación institucional a =0,991 (Excelente). Los resultados fueron analizados con del software estadístico SPSS V. 24, en donde se determinó que la gestión del cambio tiene relación positiva baja y muy significativa con la identificación institucional de los servidores públicos del Hospital Belén de Trujillo, 2018, con valor del coeficiente de Rho de Spearman 0.359 (positiva baja) y valor de Tau-b de Kendall 0.322, la significancia de correlación fue 0.002 (P<0,01); entonces se aceptó la hipótesis de investigación y se rechazó la hipótesis nula. Así mismo se identificó que el nivel de avance de la gestión del cambio de los servidores públicos del Hospital Belén de Trujillo, 2018 fue regular con 43.8%. Al igual que el nivel de la identificación institucional de los servidores públicos del Hospital Belén de Trujillo, 2018 fue regular con 41.1%

    El objetivo de este trabajo ha sido el desarrollo de animaciones para un sistema tutorial inteligente para el aprendizaje de las matemáticas para dispositivos móviles y/o tablets utilizando tecnología web dentro del proyecto EMATIC. Se pretende, por tanto, conseguir llegar al público infantil, de una manera divertida. Como resultado del trabajo se han desarrollado animaciones en la pantalla “Home” de la aplicación, en diversos escenarios y en varias actividades que antes eran estáticas

    Site-Level Web Template Extraction Based on DOM Analysis

Templates allow them to increase productivity by plugin content into already formatted and prepared pagelets. For the final user templates are also useful, because they provide uniformity and a common look and feel for all webpages. However, from the point of view of crawlers and indexers, templates are an important problem, because templates usually contain irrelevant information such as advertisements, menus, and banners. Processing and storing this information leads to a waste of resources (storage space, bandwidth, etc.). It has been measured that templates represent between 40 % and 50 % of data on the Web. Therefore, identifying templates is essential for indexing tasks. In this work we propose a novel method for automatic web template extraction that is based on similarity analysis between the DOM trees of a collection of webpages that are detected using an hyperlink analysis.     Ninia hudsoni (SERPENTES: DIPSADIDAE) as prey of the coral snake Micrurus hemprichii ortonii (SERPENTES: ELAPIDAE) in northwestern amazonia

    Objetivo: Realizar una descripción de un evento depredatorio por parte de la serpiente coral Micrurus hemprichii ortonii sobre la serpiente Ninia hudsoni. Alcance: Contribuir al conocimiento de la historia natural de serpientes neotropicales, particularmente en lo referente a interacciones tróficas. Metodología: Disección en laboratorio. Principal resultado: Este es el primer registro de interacción trófica entre M. h. ortonii y N. hudsoni. Conclusiones: La relación entre la longitud total de la presa respecto a la longitud rostro-cloaca del depredador, fue muy alta (&amp;gt; 90%), indicando que esta especie de serpiente coral puede ingerir presas de gran volumen y tamaño respecto a su propio cuerpo.Objective: To make a description of a predatory event by the Micrurus hemprichii ortonii coral snake on the Ninia hudsoni snake. Scope: Contribution to the knowledge of the natural history of Neotropical snakes, particularly in relation to trophic interactions. Methodology: Dissection in the laboratory. Main result: This is the first record of the trophic interaction between M. h. ortonii and N. hudsoni. Conclusions: The relation between the total length of the prey and the snout-vent length of the predator was very high (&amp;gt;90%), indicating that this species of coral snake can ingest preys of great volume and size in relation to its own body

    Economic Feasibility of Wireless Sensor Network-Based Service Provision in a Duopoly Setting with a Monopolist Operator

    [EN] We analyze the feasibility of providing Wireless Sensor Network-data-based services in an Internet of Things scenario from an economical point of view. The scenario has two competing service providers with their own private sensor networks, a network operator and final users. The scenario is analyzed as two games using game theory. In the first game, sensors decide to subscribe or not to the network operator to upload the collected sensing-data, based on a utility function related to the mean service time and the price charged by the operator. In the second game, users decide to subscribe or not to the sensor-data-based service of the service providers based on a Logit discrete choice model related to the quality of the data collected and the subscription price. The sinks and users subscription stages are analyzed using population games and discrete choice models, while network operator and service providers pricing stages are analyzed using optimization and Nash equilibrium concepts respectively.     TeMex: The Web Template Extractor

TeMex is fully automatic, and it can work with online webpages without any preprocessing stage (no information about the template or the associated webpages is needed) and, more importantly, it does not need a prede- fined set of webpages to perform the analysis. TeMex only needs a URL. Contrarily to previous approaches, it includes a mechanism to identify webpage candidates that share the same template. This mechanism increases both recall and precision, and it also reduces the amount of webpages loaded and processed.
